A Novel Approach to Prognostic Factors and Risk Stratification in Pediatric AML: Case Report and Literature Review - PubMed

Source : https://pubmed.ncbi.nlm.nih.gov/41096539/

Acute myeloid leukemia (AML), the second most common type of leukemia in children, is a heterogeneous disease known to be caused by genetic, epigenetic, and transcriptional changes, predominantly somatic and...

This review and case report highlight emerging genomic and molecular prognostic factors in pediatric AML, emphasizing NGS-based risk stratification, targeted therapeutic prospects, and a clinical case of high-risk M5 AML managed with stem cell transplantation.

Dual Targeting of IKKβ and NR4A1 for AML Therapy - PubMed

Dual Targeting of IKKβ and NR4A1 for AML Therapy - PubMed

IKKβ and NR4A1 are clinically relevant mediators of AML pathogenesis.A novel celastrol-based PROTAC can effectively degrade both IKKβ and NR4A1 to disrupt AML pathogenesis.

This preclinical study identifies IKKβ and NR4A1 as AML drivers and introduces a celastrol-based PROTAC (A9) that degrades both via CRBN-mediated proteasomal pathways, effectively suppressing AML progression without inducing neutrophilia.

Factors affecting response and resistance to venetoclax in acute myeloid leukemia - PubMed

Source : https://pubmed.ncbi.nlm.nih.gov/41030943/

The use of the BCL2 inhibitor venetoclax in combination with hypomethylating agents (HMA) is a revolution for the treatment of frail and elderly acute myeloid leukemia (AML) patients. This effective...

This review explores molecular and clinical determinants of venetoclax response in AML, highlighting mutation-specific sensitivity patterns, mechanisms of resistance, and implications for optimizing venetoclax-based therapeutic strategies in different patient subsets.

How to think about acute leukemia of ambiguous lineage - PubMed

How to think about acute leukemia of ambiguous lineage - PubMed

Source : https://pubmed.ncbi.nlm.nih.gov/39644014/

Classification of acute leukemia involves assigning lineage by resemblance of blasts to normal progenitor cells. This approach provides descriptive information that is useful for disease monitoring, provides clues to pathogenesis,...

Acute leukemias of ambiguous lineage (ALAL), including MPAL, pose diagnostic and therapeutic challenges due to lineage overlap. Recent classifications and genomic insights highlight disease heterogeneity, guiding evolving diagnostic and treatment strategies.
